The problem

Some microbes turn nitrogen gas from the air into ammonia, making nitrogen available to living systems. The enzyme nitrogenase carries out the conversion at a small metal-containing site. What sequence of chemical steps lets it break the bond joining the two nitrogen atoms and form ammonia?[1] [3]

Scope

Experimentally consistent molecular intermediates and reaction steps in the molybdenum nitrogenase N2-to-NH3 catalytic cycle.

What remains open

Detailed pathways are proposed, but experiments do not yet select every atomic step.[1] [2] [3]

Why it matters

This conversion connects atmospheric nitrogen to the nitrogen used in living matter. Explaining its chemistry would show how an enzyme’s metal-containing structure accomplishes a transformation that nitrogen gas does not readily undergo on its own.[1] [3]

Definitions

2 terms
active site

The region of an enzyme where the chemical reaction takes place. In molybdenum nitrogenase it includes the iron–molybdenum cofactor.[1] [3]

reaction intermediates

Temporary chemical forms between the starting material and products. A proposed intermediate still needs evidence that it belongs to the actual catalytic sequence.[1] [2] [3]
